Abstract: Bisphenol A (BPA), a ubiquitous environmental endocrine-disrupting chemical (EDC), poses significant risks to human health, particularly regarding multi-organ carcinogenesis. This presentation provides a comprehensive, mechanistic overview of how BPA drives oncogenic transformation through distinct, tissue-specific pathways, focusing on the mammary gland and the liver. In mammary carcinogenesis, while BPA predominantly acts through hormone receptor-mediated signaling during vulnerable developmental windows, recent advances reveal that it also induces profound alterations in intracellular lipid metabolism pathways. This lipid metabolic reprogramming in mammary tissues disrupts cellular homeostasis, contributing to aberrant proliferation and epigenetic remodeling. In the liver, BPA reprograms systemic metabolism and accelerates progression from metabolic dysfunction-associated steatotic liver disease (MASLD) to hepatocellular carcinoma (HCC) by inducing mitochondrial dysfunction and hepatic lipid accumulation. Furthermore, this session will highlight the critical impact of chemical co-exposures, demonstrating how BPA synergizes with dietary factors or other environmental toxins to exacerbate oncogenic signaling. By bridging these distinct yet interconnected pathophysiological routes—where lipid metabolic dysregulation serves as a common denominator in both mammary and hepatic oncogenesis—this talk aims to elucidate the multifaceted molecular landscape of BPA toxicity.
